5-Amino-1MQ is a small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT). It is stocked next to NAD-related research materials, which makes sense once the shared chemistry is clear — but it is not a peptide and not an NAD precursor. It works on the opposite side of the same molecule.

Key points
- Class: quinolinium small molecule; membrane-permeable per the originating report.
- Target: NNMT, a methyltransferase, not a receptor.
- Pathway link: intersects NAD salvage at nicotinamide.
- Literature: enzyme and cell assays, plus mouse studies; no human trial located.
- Active research area: NNMT is currently studied heavily in oncology, not only metabolism.
The one reaction that explains everything
NNMT catalyses a single transfer:
| In | Enzyme | Out |
|---|---|---|
| Nicotinamide + S-adenosylmethionine (SAM) | NNMT | 1-methylnicotinamide (MNA) + S-adenosylhomocysteine (SAH) |
Two separate consequences follow, and the literature draws on both:
- Nicotinamide is consumed. Nicotinamide is the substrate the NAD salvage pathway uses to regenerate NAD. Methylating it removes it from that pool.
- SAM is consumed, and SAH is produced. SAM is the cell’s general methyl donor, and SAH inhibits other methyltransferases. So NNMT activity also sits on the methylation economy of the cell, which is one route by which it is linked to gene regulation.
This is why NNMT appears in two otherwise unrelated bodies of research — metabolic work and cancer biology. It is the same enzyme reached from two directions.
Where it sits relative to NAD precursors
| NAD precursors (NMN, NR) | NNMT inhibition | |
|---|---|---|
| Point of action | Supply into the salvage pathway | Loss from the salvage pathway |
| Molecular class | Nucleoside / nucleotide | Small heterocycle, enzyme inhibitor |
| Secondary effects | Primarily on NAD pools | Also affects SAM / SAH methylation balance |
| Target specificity question | Which cells take up the precursor | Enzyme selectivity against other methyltransferases |
Treating these as interchangeable is the most common error in secondary descriptions. For the precursor side of the picture, see NMN compared with NAD+ and what NAD+ is.
What the published work reports
The 2018 paper by Neelakantan and colleagues in Biochemical Pharmacology is the primary reference for this compound class. It describes selective, membrane-permeable small-molecule NNMT inhibitors and reports effects in mice on a high-fat diet. Two features of that paper matter when citing it: membrane permeability was an explicit design requirement — many enzyme inhibitors fail at the cell membrane — and selectivity against other methyltransferases was addressed, which is the hard part of inhibiting an enzyme in a large family.
The more recent NNMT literature has shifted. A 2025 Nature paper reports that NNMT inhibition in cancer-associated fibroblasts restored antitumour immunity, and a 2024 study in the Journal for ImmunoTherapy of Cancer implicates fibroblast NNMT in tumour progression and immunotherapy resistance. A 2020 Journal of Hepatology study describes NNMT upregulation under ER stress in alcohol-related fatty liver. The enzyme, in other words, is a live target in several fields, and anyone reading only the 2018 metabolic paper is seeing a small slice.
Handling in the lab
- Small-molecule powder. Store sealed and desiccated at −20 °C, protected from light.
- Prepare stocks in DMSO for cell work unless the data sheet states otherwise; include a vehicle-only control, since DMSO is not inert at higher percentages.
- Aliquot stocks. DMSO is hygroscopic, and a repeatedly opened tube takes up water, changing the effective concentration.
- Confirm identity by LC-MS. Peptide methods such as sequence confirmation do not apply.
- Bacteriostatic water and peptide reconstitution tables are not relevant here — the calculator is for the peptides in a mixed study set.

Why enzyme inhibitors need different controls
A receptor ligand can often be validated by a competition experiment. An enzyme inhibitor cannot, and the controls differ accordingly. Two are worth planning for in advance:
- Substrate concentration matters. Apparent potency depends on how much nicotinamide and SAM are present, so an IC50 measured under one set of substrate concentrations will not transfer to another. Record both when comparing with a published figure.
- Product measurement beats inference. The cleanest evidence of target engagement is a fall in 1-methylnicotinamide, the reaction product. An effect observed without that measurement is consistent with NNMT inhibition but does not demonstrate it.
The same reasoning applies to selectivity. Because SAM is the shared methyl donor for a large enzyme family, a selectivity claim means a measured margin against named alternatives at the working concentration — not an absence of reported off-target effects.
Open questions
- How much of the observed effect is attributable to NAD pool changes versus SAM / SAH methylation effects?
- What is the selectivity margin against other SAM-dependent methyltransferases at working concentrations?
- Given the tissue-specific NNMT findings in tumour stroma, how do whole-animal results distribute across cell types?

Frequently asked questions
What is 5-Amino-1MQ?
5-Amino-1MQ is a small synthetic molecule, a quinolinium compound, described in the literature as a membrane-permeable inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT). It is not a peptide.
What does NNMT do?
Nicotinamide N-methyltransferase transfers a methyl group from S-adenosylmethionine to nicotinamide, producing 1-methylnicotinamide. Because nicotinamide is a substrate for NAD salvage, methylating it diverts that molecule away from NAD regeneration.
Why is NNMT studied alongside NAD compounds?
The two intersect at nicotinamide. NAD precursors supply the salvage pathway from the front; NNMT inhibition is studied as a way to reduce loss of nicotinamide from that same pathway. They are different points of intervention on one pathway.
Is 5-Amino-1MQ a peptide?
No. It is catalogued alongside research peptides but is a small heterocyclic molecule. Peptide handling guidance and reconstitution tables do not apply to it.
Has 5-Amino-1MQ been studied in humans?
No human trial was identified in our search. The published work on this compound is in enzyme assays, cell systems and mice.
